What is semaglutide?

Semaglutide is a synthetic analog of the human incretin hormone GLP-1. Endogenous GLP-1 is released from the L-cells of the intestine in response to food intake and exerts multiple glucose-dependent actions: it stimulates insulin secretion from pancreatic beta-cells, suppresses glucagon release from alpha-cells, slows gastric emptying, and promotes satiety in the central nervous system. However, native GLP-1 has a very short half-life of just 1-2 minutes due to rapid degradation by the enzyme dipeptidyl peptidase-4 (DPP-4).

Semaglutide is engineered for prolonged action. Its structure features a modification that increases its binding affinity to albumin, significantly reducing its renal clearance and making it resistant to DPP-4 degradation. This results in a half-life of approximately one week, allowing for once-weekly subcutaneous administration.
